Custom Peptide Synthesis

Custom peptides are synthesized in the laboratory utilizing the Fmoc synthesis strategy. Peptides consisting of up to 50 amino acids may be synthesized with peptides of 30 or fewer amino acids being routine. A wide range of quantities may be requested, as little as five mg or as much as one gram. In addition to the 20 normal L-amino acids, any D-amino acids or many unnatural amino acids may be incorporated into a peptide. Special modified peptides may be prepared. Examples of these modifications include N-terminal biotinylation or addition of various chromophores (Dabcyl, nitrophenylalanine, etc.), also phosphopeptides and sulfotyrosine peptides can be prepared.

The multiple antigenic peptide molecule (MAP peptide) is a macromolecule consisting of multiple copies of a desired peptide sequence. It can be prepared from virtually any amino acid sequence, although it is best to avoid cysteine residues. The MAP peptide molecule has a molecular weight ranging from 8 to 18 kDa. It consisting of a central core of branching lysine residues with multiple identical peptide chains extending outward from the core. Individual peptide subunits are attached to the central core via the C-terminal carboxyl groups. This molecule can be used as an immunogen without the need to couple it to a carrier molecule, due to its large molecular weight. The MAP peptide molecule consists of approximately 90-95% of its mass as the desired peptide sequence, with only 5-10% of the mass representing the lysine core. MAP peptides may be ordered as either 8-branch or 4-branch molecules, with each branch consisting of one copy of the desired peptide sequence. Eight-branch MAP peptides may be ordered with up to 18 amino acids in each branch, while 4-branch MAPs may contain up to 24 amino acids. MAP molecules may be ordered as crude or as HPLC "desalted", which removes any non-peptide components and any MAP molecules that are not very close to the correct structure.

Normal linear peptides may be ordered as crude or as HPLC purified material. Three levels of purity are available, 85+% pure, 90+% pure and 95+% pure. Crude peptide preparations will consist of from 50 to 80% desired peptide. The actual percentage for a given peptide will depend upon the peptide length and the particular sequence involved. Longer peptides and peptides with reactive amino acids (Cys, Met, Trp) will generally be of lower purity in the crude state. All peptides synthesized by the Facility are characterized by HPLC analysis and mass spectrometry.
